1970s
Missouri got married to her husband and had a son named James in the early 70s.
Missouri's husband had passed away at some point when her son James was very young. She became hunter following her husbands death and learned the truth about the evil in the world at some point in her life, and had gone hunting from time to time and left her hometown in her trailer. At some point in her early hunting life she befriended other hunters like Jim Murphy, Bobby Singer, Ellen Harvelle and Bill Harvelle.
She crossed paths at an unknown point in time with Mary Winchester when she was still hunting at the time. She and Mary appeared to lose touch, by the time Mary gave up hunting to settle down with John in Lawrence, while Missouri kept hunting and hadn't been in Lawrence for seven years by 1983.
1983
In 'The Hunt Has Begun' during flashbacks, Missouri reads in a newspaper about the death of Mary Winchester in a house fire, which prompts her to return to Lawrence, she discusses the news with other hunters when she calls her friend Ellen Harvelle at the Roadhouse, learning it is the sixth house fire death in the past year.
Missouri drives her trailer back into her old hometown and approaches the grieving John Winchester in a bar, where she reveals to him that there is evil in the world, and that is what killed Mary.
John returned to Missouri's trailer a week later where Missouri told him more about demons and monsters and also confessing that his wife was a hunter like her, although she lost touch with Mary for several years, as Missouri had been hunting on the road for several years driving around in her trailer. She tells John she has a son about the same age as his son Dean. Missouri helped John to find out more about what might have killed Mary, and the two follow signs by Missouri's visions and enter a house, only to find it was really a a diversion to get to Dean and Sam .Which prompts Missouri to race back to his house with John, to find the boys fine, but both his sister and brother and law dead.
Missouri sets John on a path to visit her friend Jim at his church. She helps John and Jim perform a spell to try and learn more about Mary's death, and she sets John on his path to become a hunter.
Between 1984 and 2005
Missouri kept in touch with John over the years, although she eventually becomes a semi-retired hunter and moved back to Lawrence at some point, and continued to use her psychic gifts to help people in her home business.
Season One
In 'Home' Sam and Dean return to Lawrence following a premonition that Sam had about their old home being haunted again. They search the phone directory for psychics, and Dean stumbles upon her and quickly remembers her from his childhood, and they go to see her.
Missouri recognizes Sam and Dean immediately when they meet her, and as she helps Sam and Dean with a ghost problem at their old house and identifies the spirit as a poltergeist. With the help of Missouri, Sam and Dean try to perform a ritual to banish the poltergeist which has taken up residence in the Winchesters' old home. However, it is Mary's spirit that eventually succeeds in defeating the poltergeist. After the poltergeist is gone, Missouri tells the boys "Don't be strangers!"
Near the end of the episode, John has been secretly at Missouri's house the whole time his boys were in Kansas, but has reluctantly kept himself hidden. Missouri helps John learn the truth using her gifts.
In 'No Exit' John had been staying in Lawrence to learn more, with the help of Missouri and also Jim. Missouri told John that Sam is a very special child. She convinces him that he needs to tell them the truth, but John again tells her not just yet. Missouri rolls her eyes, he better grow a pair and do it soon, or she will.
In 'Darkness Calling', Missouri had been working with John and Jim to find more special children, the demon and the Colt. With Missouri continuing to be annoyed that John was lying to his boys.
In 'Faith' and 'The Winchester Reunion' Missouri helps Dean, Sam, Jo and Jim with a case close to home, dealing with a Faith healer, and Reapers.
Missouri convinces John to return to Lawrence to see his boys, and he shocks everyone including Missouri when he shows up at her door step. Missouri is overjoyed that John has finally told his boys the truths, and together she and John tell Dean, Sam and Jo about the Colt. She promises to keep looking for information and clues to find the Colt, when she everybody leaves her home.
In
'The Life and Death of Daniel Elkins' she came to pay her respects and attended the Hunters wake for Daniel Elkins at the Roadhouse. She mentioned at the wake how impressed she was that he had a good turnout with lots of hunters attending. She was pleased to hear that John had kept the Colt hidden well.
Missouri got a vision of an incident that would occur at the Original Roadhouse in Nebraska when she touched Jo's shoulder, but kept it to herself, as the best outcome to save everybody was to plan a Devil's Trap herself. Which would happen in a few days time.
Later, when the group gets worried about John and Jim working a case with the La Mala Hora. Missouri altered them on a vision prompting herself, Dean and Sam to head to New Mexico. They manage to save John and Jim. Not before learning that John lied about keeping the Colt safe, when he used a bullet from the Colt to save Dean's life.
After the case Missouri headed back to Lawrence and told the Winchesters she'll continue to dig into the whereabouts of Yellow Eyes or his children
In
'Salvation' Missouri is the one to tragically find Jim bleeding to death when he was killed by Meg the demon. She mourns the loss of her friend,
wishing she could have helped him, and that she didn't even get a vision of this. Jim proceeds it wasn't her fault, and admits that back in New Mexico, he looked at the La Mala Hora eyes briefly, and likely got cursed and was destined to die anyway. Jim begged Missouri to leave town immediately, as Meg will be looking for her to get John's location out of her.
Missouri informed John over the phone of Jim's death. Revealing that she found traces of sulfur at Jim's place.
In
'Devil's Trap (Part 1 and 2)' Missouri was contacted by Ellen who was concerned about not hearing from her
after she agreed to leave Kansas for her safety. But to their surprise Missouri had already on her way to the roadhouse.
Missouri enters the Roadhouse finding the door is already open and doesn't find Bobby or Ash inside, but instead finds that a demon had been there, when she finds sulfor. She finds Ash's phone, and finds a message on it "Nebraska. The Original HR". She tells them about the message. Ellen immediately knew what it meant, which was The Original Harvelle Roadhouse in Nebraska which was where Bill was born, and the place where Bill was killed at on a hunt with John.
Dean and Sam arrive at the Roadhouse, and together with Missouri researched some of Bobby and Ash's research and Bobby's books on demon lore. She gives Sam the Key of Solomon. When Meg turns up at the Roadhouse, they restrain her using a devil's trap (symbol) painted on the ceiling. Missouri then alerts the boys to the fact that Meg is not a demon herself, but a woman possessed by one, which John suspected before. After the demon is exorcised, and and the real Meg returns, she thanks them for freeing her and offers the clue "sunrise". Missouri wanting to make sure Meg is safe, sends her to another old friend of Bobby's also a hunter who had gone in retirement, who will check up on Meg's sister, and make sure it is safe to go home.
Later, Missouri headed to the O.G Roadhouse in Nebraska, to save and rescue Bobby, Ellen and Jo and stop the "Hellspawn" that had been taking possession of Ash. She revealed the truth about her vision at Daniel's wake. Together they perform a ritual, which is different than a regular demon possession one. Which requires all of them to channel each other, and they'll use their own energy to pull the hellspawn out of Ash, but if any of them break the link they form, it might cause consequences for Ash.
They manage to finish the ritual, and when the true form of the Hellspawn shows itself, they all manage to kill it.
Missouri later contacted the Winchesters concerned about not hearing from him. She learned that they faced the Yellow Eyed Demon, survived, but are heading to the hospital.
They receive a call from Missouri who asks if they are okay, concerned when she didn't hear back from them. Missouri is horrified to hear they faced the Yellow Eyed Demon, survived but are all badly hurt. Missouri was promised a call when they reached the hospital, but said call never came because The Professor smashed the Impala leaving them critically injured.
