Charlie said goodbye to Sam when he left town, she understood that he needed time away, due to Jessica's death.
A few months later. in 'Skin'. Charlie sent Sam an email asking for his help when her girlfriend had been arrested for the murder of their friend. Sam and Dean came to St. Louis to help. By Slumber Party, Charlie had become a full-fledged hunter, having completed cases of her own but only as a hobby. Her cases included those involving a ghost and a teenage vampire. Despite being a part-time hunter, she felt her life was incomplete and wanted to find a quest of her own. Charlie was called in by Sam and Dean to use an ancient computer they found in an effort to find all of the angels in the world. Charlie hacked the computer and downloaded all of the Men of Letters files, but later came into conflict with The Wicked Witch when she was accidentally released by Dean. Charlie worked with Dorothy to hunt the witch, interested in Dorothy from the Oz books she read as a child. Charlie was killed by the Witch, but Dean has Gadreel (in the guise of Ezekiel) resurrected her. Dorothy claims that Charlie went to Heaven. Charlie came up with the idea to use poppy extract bullets to stun the Witch and realized that the books were written by L. Frank Baum to give Dorothy clues on how to deal with the Witch.
Thanks to Charlie's revelation, Dorothy realized that the Ruby Slippers can be used to kill the Witch. While Dorothy holds off a possessed Sam and Dean, Charlie goes after the Witch and supposedly killed her with the Slippers just as she was about to summon her army to Earth. Charlie manages to shut the door to Oz just in time,

Upon reaching Oz, Charlie and Dorothy managed to put down the Witch's rebellion. Once peace was achieve, Dorothy asked Charlie to remain in Oz, to help the transition to peace. Charlie agreed.
As time passed, Charlie and Dorothy slowly fell in love, and were married.
Dorothy Baum, the love of Charlie's life.
About a year or so later, the couple discovered that the Wicked Witch was still alive and had formed an alliance with the Queen Of Hearts, who lived in the neighbouring realm of Wonderland. The two evil beings then cast a spell on the solidiers of both Oz and Wonderland, hoping to start a way between the two lands. In the aftermath of said war, the Witch and Queen would set themselves up as dictators of a totalitarian state and rule over the survivors of both lands.
To prevent this, Dorothy sent Charlie back to Earth, to collect Dean and Sam, knowing that they could help. The three hunters confronted the evil pair and managed to defeat them. The Witch and Queen ended up banished to an empty dimension, where they could cause no further harm. With the evil pair gone, the spell was broken and peace between Oz and Wonderland was restored.
Later, back in Emerald City, Dorothy thanked Dean and Sam for their help, and opened a portal for them to return to Earth. When Dean asked if Charlie intended on returning as well, she said no. Charlie revealed that she and Dorothy are now married and that Oz is her home now. Before leaving for Earth, Dean and Sam congratulated the pair, happy that Charlie has at last found true love with Dorothy.
